The Assay Genie KIR3DL1 immunoassay is a quantitative sandwich ELISA for measuring soluble Killer cell immunoglobulin-like receptor 3DL1 in human serum and plasma samples. KIR3DL1 is a key regulatory receptor expressed on natural killer (NK) cells and a subset of CD8+ T lymphocytes, where it surveys MHC class I molecules bearing the HLA-Bw4 motif and modulates cytolytic activity. Researchers apply this microplate assay across immunology, transplantation, and infectious disease studies, including NK cell education, reproductive immunology, and KIR-HLA interactions in HIV control and HCV outcomes.

The microplate is pre-coated with a human-specific capture antibody that shows no significant cross-reactivity with mouse or rat KIR3DL1, allowing the receptor's extracellular domain to be bound from biological matrix before detection with a biotinylated secondary antibody and an HRP-streptavidin conjugate. A standard curve is generated using recombinant KIR3DL1 protein supplied in the kit, and the substrate yields a colorimetric readout proportional to receptor concentration. Each kit includes reagents for one 96-well microplate: a coated plate, lyophilized standards, detection antibody, streptavidin-HRP, wash buffer, substrate solution, and stop solution. Reconstituted components should be refrigerated at 2-8°C; the sealed plate and lyophilized standards remain stable until the printed expiration when stored at 2-8°C, and repeated freeze-thaw cycles of the standards should be avoided. This kit is supplied for research use only, for in vitro applications, and is not intended for diagnostic procedures.
