The Assay Genie ELISA-format quantitative immunoassay measures human KIR2DL3, also known as CD158b2 or NKAT2, a Type I transmembrane inhibitory receptor belonging to the Killer-cell Immunoglobulin-like Receptor family. Expressed on natural killer cells and a subset of CD8-positive T lymphocytes, KIR2DL3 contributes to NK cell education and modulates cytotoxic responses against target cells displaying cognate HLA-class I molecules.

KIR2DL3 specifically engages HLA-C group 1 allotypes, defined by an asparagine residue at position 80 of the heavy chain, in contrast to the related inhibitory receptor KIR2DL1, which recognizes group 2 allotypes carrying lysine at that position. Researchers apply this kit to studies of NK cell repertoire diversity, viral immunity (including the well-characterized influence of KIR2DL3 in combination with HLA-C1 ligands on HIV-1 disease progression), reproductive immunology in recurrent pregnancy loss, donor-recipient matching in hematopoietic stem cell transplantation, and broader investigations of immune regulation in inflammatory and autoimmune contexts.

Each kit supplies a pre-coated microplate, biotinylated detection reagent, enzyme conjugate, reference standards, and buffer reagents in a microplate format suitable for batch processing. The workflow follows standard incubation and wash steps with a chromogenic readout on a conventional plate reader. Components should be refrigerated between uses and protected from light; full reconstitution volumes, suggested dilution factors for serum-derived or plasma-derived research specimens, and assay-specific calibration guidance are detailed in the accompanying manual.
