Description

A sandwich ELISA kit from Assay Genie, designed for the quantitative measurement of human Ketohexokinase (KHK), also known as hepatic fructokinase. The enzyme catalyzes the phosphorylation of the ketose sugar fructose to fructose-1-phosphate and represents the first enzyme with a specialized pathway that catabolizes dietary fructose. KHK plays a critical role in fructose metabolism and has been associated in the literature with metabolic disorders, diabetes and liver diseases, supporting research into the mechanisms of these conditions and the identification of potential therapeutic targets.

The assay is specific for natural and recombinant human Ketohexokinase and is intended for use with human samples, including serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. Reported performance characteristics include a detection range of 0.312-20ng/mL, a sensitivity of 0.078ng/mL, an intra-assay CV of 4.9% and an inter-assay CV of 8.6%. Linearity has been demonstrated in serum, EDTA plasma and heparin plasma across dilutions of 1:2 to 1:16, with reported recovery averaging 93% in serum (range 87-99%) and 95% in plasma (range 89-101%). The target protein is a homodimer with UniProt accession P50053 and a molecular weight of 32,730 Da. The kit is for research use only.

Detection is carried out using a standard sandwich ELISA workflow, with absorbance read at 450 nm on a microplate reader after the addition of substrate and stop solution. Detailed reagent preparation, incubation times, wash steps and sample-handling guidance should be taken from the protocol supplied with the specific kit lot.
