JPH1 (Junctophilin-1) is a membrane-binding structural protein of the junctional membrane complex, anchoring the sarcoplasmic reticulum to transverse-tubule plasma membrane in striated muscle and supporting excitation-contraction coupling. Sequence variants in JPH1 have been associated with the neuromuscular disorder Charcot-Marie-Tooth disease type 2K, while altered JPH1 expression has been reported in studies of Duchenne muscular dystrophy and other skeletal muscle pathologies.

This Assay Genie quantitative immunoassay measures human Junctophilin-1 levels in serum biospecimens and in plasma samples prepared with standard anticoagulants, with a colorimetric readout captured on a standard microplate reader. The kit ships as a 96 Assays microplate configuration, is validated for human reactivity, and is intended for research use only in academic and industry laboratories studying muscle biology, neuromuscular disease biomarkers, and excitation-contraction coupling mechanisms.

Each kit supplies a pre-coated 96 Assays microplate, biotinylated detection antibody, recombinant Junctophilin-1 calibrator standards, streptavidin-HRP conjugate, wash buffer concentrate, TMB substrate, stop solution, and assay diluent sufficient to run a full plate. Store kit components refrigerated between uses and protect the TMB substrate from light. Aliquot reconstituted standards and avoid repeated freeze-thaw cycles of working reagents.

Typical applications include biomarker screening in neuromuscular disease cohorts, characterization of skeletal muscle cell lines and primary myotube cultures, evaluation of contraction models, and pharmacodynamic studies of compounds influencing junctional membrane integrity in striated muscle.
