Description

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of human Isocitrate dehydrogenase [NADP] cytoplasmic (IDH1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay uses a colorimetric detection method with reactivity restricted to human samples and recognises both natural and recombinant forms of the target protein. The research area indicated by the manufacturer is cardiovascular, and the kit is supplied for research use only.

IDH1 is an oxidoreductase that catalyses the third step of the TCA cycle, converting isocitrate to α-ketoglutarate while consuming NADP(+). It functions as a homodimer located in the cytoplasm and peroxisome, and the associated UniProt entry is O75874. Mutations in the IDH1 gene, particularly at arginine 132 (R132), have been found in 80% of grade II–III gliomas and secondary glioblastomas, and neomorphic IDH1/IDH2 mutations occur in a high percentage of patients with cytogenetically normal acute myeloid leukaemia, making the enzyme a relevant biomarker in oncology and metabolism research.

According to the manufacturer's specifications, the detection range is 0.78-50ng/mL and the sensitivity is 0.31ng/mL. Reported precision is 5.8% intra-assay CV and 9.9% inter-assay CV. Linearity has been validated in serum, EDTA plasma and heparin plasma across dilutions from 1:2 to 1:16. Recovery is 98% (range 92-104%) in serum and 100% (range 94-106%) in plasma.
