IDH1 is a cytosolic NADP-dependent enzyme that catalyzes the oxidative decarboxylation of isocitrate to alpha-ketoglutarate while reducing NADP+ to NADPH. The encoded protein runs at approximately 46 kDa. Mutations in IDH1, particularly the R132H hotspot, are frequently observed in gliomas and acute myeloid leukemia, where they drive production of the oncometabolite 2-hydroxyglutarate and alter epigenetic regulation through effects on histone and DNA methylation. Quantitative measurement of soluble IDH1 protein supports cancer metabolism research, biomarker discovery, and investigations into cellular redox homeostasis. This immunoassay from Assay Genie is configured for human specimens and accepts starting material including plasma specimens, serum specimens, tissue homogenates, cell lysates, and cell culture supernatants. The kit is supplied in a microplate format and uses an antibody-pair capture and detection approach with absorbance readout on a standard plate reader. Researchers investigating glioma biology, leukemia metabolism, or the downstream consequences of mutant IDH1 can apply the kit to patient-derived tissue, xenograft samples, and relevant cell line models. For research use only; not intended for diagnostic applications.
