Human IRF5 ELISA Kit

From Assay Genie, this sandwich ELISA is designed for the precise quantification of IRF5 (Interferon Regulatory Factor 5) in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. IRF5 is a key regulatory protein involved in the immune response, with dysregulation linked to autoimmune diseases such as lupus and rheumatoid arthritis, making it a relevant biomarker for immunology research.

The assay uses a colorimetric readout with a sensitivity of 9.375 pg/mL across a standard range of 15.625–1000 pg/mL. Reported linearity at 1:2, 1:4 and 1:8 dilutions is 89–99% / 82–97% / 80–91% for serum, 86–93% / 90–99% / 91–96% for EDTA plasma, and 90–105% / 85–100% / 82–99% for heparin plasma (n = 5). Recovery ranges are 86–96% for serum, 88–101% for EDTA plasma and 88–105% for heparin plasma, with average recoveries of 92%, 95% and 95% respectively. Unopened kit components are stored at 2–8 °C and remain stable for 12 months under those conditions.

The workflow uses a pre-coated microplate: standards, samples and controls are incubated for 90 minutes at 37 °C, followed by a 60-minute biotin-labelled detection antibody step, a 30-minute HRP-Streptavidin (SABC) binding step, and TMB substrate development in the dark for 10–20 minutes, with absorbance read at 450 nm after stop solution is added. Components supplied include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, 25X wash buffer, plate sealer and a technical manual.
