Description

Manufactured by Assay Genie, this Sandwich ELISA Kit (Double Antibody format) is designed for the quantitative detection of IRE1 in human samples. The assay is reactive with Human specimens and accepts a broad range of sample types including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit delivers a sensitivity of 37.5 pg/mL with an analytical range of 62.5-4000 pg/mL and is read colorimetrically using a TMB substrate, with absorbance measured at 450 nm after stop solution addition.

Linearity has been validated at 1:2, 1:4 and 1:8 dilutions across serum (n = 5) at 86-101%, 88-96% and 81-97%; EDTA plasma (n = 5) at 94-104%, 87-100% and 84-96%; and heparin plasma (n = 5) at 85-98%, 84-99% and 82-97%. Recovery ranges reported are 87-98% (average 92%) for serum (n = 5), 90-102% (average 98%) for EDTA plasma (n = 5) and 97-101% (average 99%) for heparin plasma (n = 5). The assay protocol involves primary incubation at 37°C for 90 minutes, detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, and color development in the dark for 10-20 minutes. Components are stored at 2-8°C (with light-protected items kept away from direct light), and shelf life is stated as 12 months under these conditions.

IRE1 (UniProt O75460; synonyms include ERN1, IRE1, IRE1a, Ire1-alpha and hIRE1p) is a key component of the unfolded protein response pathway, regulating cellular stress responses and maintaining cellular homeostasis. Its dysregulation has been implicated in cancer, diabetes and neurodegenerative disorders, making the kit a relevant tool for research in the Cell Death area, including disease pathogenesis studies and biomarker investigations.
