Description

A colorimetric sandwich ELISA kit from Assay Genie is designed for the quantitative detection of human IPAF/NLRC4. Reactivity is specific to human samples, with validated use on serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Performance characteristics include a sensitivity of 46.875 pg/mL and an assay range of 78.125-5000 pg/mL. Detection is based on a double-antibody sandwich format with TMB substrate development, and absorbance is read at 450 nm.

The assay workflow consists of reagent and plate preparation, a 90-minute primary incubation at 37°C, biotin-labeled detection antibody binding for 60 minutes at 37°C, HRP-Streptavidin binding for 30 minutes at 37°C, TMB color development in the dark for 10-20 minutes, and a stop step followed by absorbance measurement at 450 nm. Reagents should be stored at 2-8°C for 12 months. The kit targets NLRC4 (Uniprot Q9NPP4), with recognised synonyms including NLR family CARD domain-containing protein 4, Ice protease-activating factor, Ipaf, CARD, LRR, and NACHT-containing protein, Clan protein, and Caspase recruitment domain-containing protein 12. The indicated research area is Cell Death.

Reported linearity in serum (n=5) across 1:2, 1:4, and 1:8 dilutions was 89-98%, 85-103%, and 88-105% respectively; for EDTA plasma (n=5) it was 83-94%, 83-99%, and 83-101%; and for heparin plasma (n=5) it was 80-96%, 85-100%, and 82-94%. Spike-and-recovery values were 87-98% (average 92%) for serum (n=5), 87-102% (average 95%) for EDTA plasma (n=5), and 86-104% (average 96%) for heparin plasma (n=5).
