Description
From Assay Genie, this sandwich ELISA kit enables the quantitative measurement of human Involucrin (INV), a key protein involved in keratinization and the formation and maintenance of the skin barrier. Designed for research applications in cell biology, the kit supports investigations into skin physiology and pathology, where changes in involucrin levels have been linked to various skin diseases and conditions.
The assay uses a colorimetric double-antibody sandwich format with a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL. It is reactive with human samples and accepts a range of liquid specimen types, including serum, plasma, cell culture supernatant, and cell or tissue lysate. Validated linearity across serum, EDTA plasma, and heparin plasma dilutions (1:2, 1:4, and 1:8) falls between approximately 82% and 104%, and recovery in the same matrices ranges from 89% to 105%.
The protocol follows a standard sandwich workflow: equilibrate reagents and TMB substrate to room temperature, load standards and samples onto the pre-coated plate for a 90-minute incubation at 37°C, add a biotin-labelled detection antibody for 60 minutes, apply HRP-Streptavidin for 30 minutes, develop colour in the dark for 10–20 minutes, stop the reaction, and read absorbance at 450 nm. Reagents are stored at 2–8°C for 12 months. The target protein is also known by the synonym IVL and corresponds to UniProt entry P07476.
