The alpha subunit of the interleukin-3 receptor, IL-3Rα, is also known as CD123 and serves as a clinically relevant biomarker in acute myeloid leukemia, blastic plasmacytoid dendritic cell neoplasm (BPDCN), and allergic inflammation studies. This Assay Genie sandwich ELISA employs a human-specific capture antibody to enable quantitative detection of soluble IL-3Rα across multiple specimen matrices, including tissue homogenates, with minimal cross-reactivity to mouse or rat homologues. The microplate protocol uses a 100 uL sample volume per well, with a recombinant standard curve included for interpolating unknown concentrations. Applications include preclinical evaluation of CD123-directed therapeutics such as tagraxofusp (SL-401), investigation of basophil and plasmacytoid dendritic cell activation, and biomarker monitoring in translational hematology workflows.
