Description

From Assay Genie, this Sandwich-format ELISA kit is designed for the quantitative measurement of human Interferon regulatory factor 8 (IRF8) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The kit uses a colorimetric detection method, is reactive with human samples and is intended for research use only.

Reported performance characteristics include a detection range of 31.2-2000pg/mL and a sensitivity of 15.61pg/mL. Intra-assay CV is 6.8% and inter-assay CV is 8.4%. Linearity was tested at 1:2, 1:4, 1:8 and 1:16 dilutions across serum (N=5), EDTA plasma (N=5) and heparin plasma (N=5), with recovery averaging 104% for serum (range 98-110%) and 106% for plasma (range 100-112%). The assay shows specificity for natural and recombinant human IRF8 (UniProt Q02556).

IRF8 is a transcription factor of the interferon regulatory factor family. It binds the interferon consensus sequence (ICS) upstream of type I IFN and IFN-inducible MHC class I genes, plays a negative regulatory role in immune cells, and participates in CD8(+) dendritic cell differentiation via a BATF-JUNB heterodimer. The protein is predominantly expressed in lymphoid tissues, localises to the cytoplasm in resting macrophages and translocates to the nucleus upon IFN-gamma induction. Dysregulation of IRF8 has been implicated in autoimmune diseases, inflammatory disorders and cancer, making it a target of interest in immunology research.
