Description

Assay Genie's colorimetric sandwich ELISA kit is designed for the quantitative measurement of Human Insulin (INS) in human samples, using highly specific antibodies pre-coated on a microtiter plate. The assay is compatible with routine laboratory and high-throughput screening (HTS) formats.

The workflow uses a quantitative sandwich ELISA technique. Standards or samples are added to microplate wells pre-coated with an antibody specific to Human Insulin (INS), followed by sequential addition of a biotinylated detection antibody specific for Human Insulin (INS) and an Avidin-Horseradish Peroxidase (HRP) conjugate. After washing to remove non-specific binding, a substrate solution is added and the enzyme–substrate reaction is terminated by the addition of a sulphuric acid solution. Optical density is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm, with the OD value proportional to the concentration of Human Insulin (INS) in the samples when compared to the standard curve.

Performance characteristics include an assay range of 1.56-100 mIU/L and a sensitivity of 0.78 mIU/L. The assay type is sandwich, with an Intra-Assay CV of 4.3% and an Inter-Assay CV of 7.5%. Compatible sample types include serum, plasma, saliva, urine, cell culture supernatant, tissue samples and other related supernatants. For long-term storage, the whole kit (excluding the substrate) should be kept at -20 °C, and the kit may be used throughout its expiration date of six months from the date of manufacture when stored as prescribed. The kit is intended for Research Use Only and includes an ultra-fast protocol designed to deliver results rapidly.
