Description

A high-sensitivity sandwich (double antibody) ELISA from Assay Genie designed for the quantitative measurement of human INPP5D, also known as SHIP1 (Phosphatidylinositol-3,4,5-trisphosphate 5-phosphatase 1; UniProt Q92835). The kit is for research use only and detects the target in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Detection is colorimetric: HRP-catalysed TMB substrate produces a blue product that turns yellow on addition of stop solution, and absorbance is read at 450 nm. The kit also includes a validated Bradford Reagent for total protein quantification to support sample normalisation.

Performance characteristics include a measuring range of 78.125–5000 pg/mL and a sensitivity of 46.875 pg/mL. The protocol consists of equilibrating reagents to room temperature, loading standards, samples and blanks onto the pre-coated plate, and incubating at 37 °C for 90 minutes to allow antigen binding. A biotin-labelled detection antibody is then added and incubated at 37 °C for 60 minutes, followed by HRP-Streptavidin (SABC) for 30 minutes at 37 °C, TMB substrate development in the dark for 10–20 minutes, stop solution, and immediate absorbance measurement at 450 nm.

Reported linearity (n = 5) at 1:2, 1:4 and 1:8 dilutions is 81–105%, 81–101% and 85–100% for serum; 80–105%, 80–101% and 80–94% for EDTA plasma; and 82–103%, 82–101% and 85–100% for heparin plasma. Recovery ranges and means are 81–90% (mean 85%) for serum, 82–96% (mean 86%) for EDTA plasma and 80–94% (mean 86%) for heparin plasma. Reagents are stored at 2–8 °C, with the kit stable for up to 12 months under these conditions.
