Description

A colorimetric sandwich ELISA designed for the quantitative measurement of inducible nitric oxide synthase (iNOS) in human samples. Produced by Assay Genie, the kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Detection is based on a double-antibody sandwich format with a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. The assay is read at 450 nm following TMB substrate development, and components are intended to be stored at 2–8°C for up to 12 months. Inducible nitric oxide synthase is a key enzyme in nitric oxide signalling, and its dysregulation has been implicated in inflammation, neurodegenerative disorders and cancer, making accurate quantification valuable for cell biology research. The target corresponds to UniProt entry P35228.

The assay workflow includes reagent and plate preparation at room temperature, a 90-minute primary incubation at 37 °C, a 60-minute biotin-labelled detection antibody step, a 30-minute HRP-streptavidin binding step, and 10–20 minutes of TMB colour development in the dark before stop solution is added and absorbance measured at 450 nm. Reported linearity across 1:2 to 1:8 dilutions is 95–105% for serum, 86–100% for EDTA plasma and 87–93% for heparin plasma. Spike-recovery averages are 93% in serum, 95% in EDTA plasma and 99% in heparin plasma. Sample-handling guidance covers serum (clot, centrifuge 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge 1000 × g for 15 minutes at 2–8 °C), tissue homogenate, cell culture supernatant and cell lysate preparations.
