The Assay Genie IκBα ELISA supports pathway-kinetics studies in which canonical NF-κB activation is monitored through stimulus-coupled IκBα turnover. In practice, lysates from HeLa, THP-1, HEK293, or Jurkat cells are harvested after short treatments with TNF-α, LPS, IL-1β, or PMA, and the cytosolic fraction is then assayed alongside unstimulated controls to quantify degradation kinetics.
Endogenous IκBα (NFKBIA, UniProt P25963) is a ~36 kDa ankyrin-repeat inhibitor whose proteasomal destruction is gated by IKK-mediated phosphorylation at Ser32 and Ser36. For lysate input, 10 to 50 µg of total protein per reaction is a common working range following BCA or Bradford normalization, and the complete workflow is typically finished in roughly 3 to 4 hours at room temperature. The microplate and lyophilized standards are stored at -20 °C between runs, while the wash buffer is held at room temperature; reconstituted standards should be aliquoted and frozen immediately to limit freeze-thaw loss.
