Description

This sandwich double-antibody ELISA kit from Assay Genie is designed for the quantitative measurement of human Ischemia Modified Albumin (IMA). It is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The microplate is pre-coated with a capture antibody specific to the target protein, and a biotinylated detection antibody binds a separate epitope to form a sandwich complex. Detection is achieved with HRP-conjugated Streptavidin and a TMB substrate, with absorbance read at 450 nm using a microplate reader. The signal intensity is directly proportional to analyte concentration, as determined from a standard curve.

Reported analytical specifications include a sensitivity of 4.688 ng/mL and a measurable range of 7.813-500 ng/mL. Spiked recovery in serum averaged 97% (range 92-105%, n=5), in EDTA plasma averaged 93% (range 86-99%, n=5), and in heparin plasma averaged 93% (range 87-105%, n=5). Dilutional linearity in serum (n=5) was 85-99% at 1:2, 82-96% at 1:4, and 82-95% at 1:8; EDTA plasma gave 83-97%, 80-99%, and 82-95% at the same dilutions, while heparin plasma gave 82-98%, 80-100%, and 83-97%. The kit is stored at 2-8°C for 12 months.

The assay workflow involves equilibrating reagents, loading standards and samples for a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody incubation, a 30-minute HRP-Streptavidin incubation, TMB color development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stopping the reaction. Serum samples are clotted and centrifuged at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2-8°C.
