Human IL-6 ELISpot Pair

The ELISpot Pair from Assay Genie supplies matched capture and biotinylated detection antibodies for enumerating human IL-6 secreting cells at single-cell resolution. This pair-based configuration lets researchers optimize coating concentration, plate membrane, and cell input for their stimulation protocol. In a standard workflow, a PVDF-bottomed plate is activated with ethanol, coated overnight with the capture antibody, blocked, and then loaded with stimulated peripheral blood mononuclear cells (PBMCs) at 100,000 to 500,000 cells per well in 100-200 µL of culture medium. After 16-24 hours of incubation, secreted IL-6 is captured in situ and visualized as discrete spots using a streptavidin-enzyme conjugate and precipitating substrate, with each spot representing a single secreting cell. Applications include vaccine immunogenicity studies, T cell functional profiling, inflammatory disease research, and monitoring cytokine responses in infectious disease models. Kit components should be stored refrigerated, with reconstituted detection antibody reserved for limited freeze-thaw cycles; the reagents are research use only and not intended for diagnostic procedures.
