Human IL-4 ELISpot Kit

This ELISpot kit from Assay Genie enables single-cell detection of IL-4 secreting cells, providing a quantitative measure of cytokine-producing immune cells. IL-4 is a key Th2-type cytokine involved in B cell class switching, humoral immunity, and allergic inflammatory responses, making this assay relevant for vaccine development, allergy research, and T cell functional studies.

The microplate-based workflow includes pre-coated membrane wells, biotinylated detection antibody, streptavidin-enzyme conjugate, AEC substrate solution, and wash buffer. Following cell stimulation and incubation, secreted IL-4 is captured locally, producing discrete spots that correspond to individual secreting cells. Spots can be quantified using an automated reader or counted manually under a dissection microscope.

Store all unopened reagents at 4°C; protect the substrate from light. Reconstituted detection antibody should be aliquoted to limit repeated freeze-thaw cycles. The assay is intended for research use only and supports in vitro studies of human peripheral blood mononuclear cells (PBMCs) and isolated lymphocyte populations from human donors.
