Human IL-32 ELISA Kit (HUFI01624)

This Sandwich ELISA from Assay Genie is designed for the quantitative measurement of human Interleukin-32 (IL-32). Using a colorimetric, double-antibody sandwich format, the kit detects IL-32 with a sensitivity of 9.375 pg/mL across a working range of 15.625–1000 pg/mL and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The assay protocol involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks onto the pre-coated plate, then incubating for 90 minutes at 37 °C. This is followed by addition of a biotinylated detection antibody with a 60-minute incubation, HRP-Streptavidin binding for 30 minutes, TMB color development for 10–20 minutes in the dark, and absorbance measurement at 450 nm after stop solution is added. Reagents are stored at 2–8 °C, with the kit stable for 12 months under these conditions.

Linearity has been demonstrated across serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, with recovery ranging from 90–105% (average 94%) in serum, 86–99% (average 94%) in EDTA plasma, and 89–96% (average 96%) in heparin plasma. Interleukin-32 is a key secreted cytokine involved in inflammation and immune response regulation, with dysregulation linked to inflammatory and autoimmune conditions.
