Human IL-26 ELISA Kit

This Sandwich ELISA from Assay Genie is designed for the quantitative measurement of human Interleukin-26 (IL-26), also known as Protein AK155. IL-26 is a cytokine involved in immune and inflammatory responses and has been implicated in inflammatory skin disorders, inflammatory bowel disease and arthritis. The kit uses a double-antibody sandwich format with colorimetric detection and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, offering a sensitivity of 9.375 pg/mL and an analytical range of 15.625–1000 pg/mL.

The procedure follows a six-step protocol: equilibrate reagents and TMB substrate to room temperature before loading standards, samples and controls onto the pre-coated plate and incubating at 37°C for 90 minutes; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate for 10–20 minutes in the dark; then add stop solution and read absorbance at 450 nm immediately.

Sample preparation guidance includes allowing blood to clot and centrifuging serum at 1000 × g for 20 minutes; collecting plasma in anticoagulant tubes and centrifuging at 1000 × g for 15 minutes at 2–8°C; homogenising tissue in PBS with protease inhibitors; centrifuging cell culture supernatant at 2500 rpm for 5 minutes; and lysing cells using lysis buffer with protease inhibitors. Spiked-recovery averages were 98% in serum, 92% in EDTA plasma and 95% in heparin plasma. Unopened kit components should be stored at 2-8°C for up to 12 months.
