Description

A Sandwich ELISA Kit from Assay Genie designed for the quantitative measurement of human Interleukin-21 (IL-21) in research samples. The assay uses a colorimetric, double-antibody sandwich detection method and is suitable for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with reactivity for human specimens. The kit is intended for cell biology research applications.

Performance specifications include a sensitivity of 37.5 pg/mL and a standard curve range of 62.5-4000 pg/mL. Reported linearity at 1:2, 1:4, and 1:8 dilutions is 87-104%, 87-90%, and 87-105% for serum (n=5); 86-100%, 82-96%, and 83-101% for EDTA plasma (n=5); and 85-99%, 84-95%, and 80-94% for heparin plasma (n=5). Recovery ranges are 88-105% (average 94%) for serum, 87-105% (average 100%) for EDTA plasma, and 90-102% (average 98%) for heparin plasma. Reagents are stored at 2-8°C for up to 12 months. The procedure involves a 90-minute primary incubation at 37°C, a 60-minute biotin-labeled detection antibody incubation, a 30-minute HRP-Streptavidin (SABC) incubation, TMB substrate development in the dark for 10-20 minutes, and absorbance measurement at 450 nm.

Interleukin-21 is a cytokine involved in the regulation of immune responses, particularly in T cell and B cell development. Dysregulation of IL-21 has been implicated in autoimmune diseases, inflammatory disorders, and cancer, making it a useful biomarker for studying these conditions and potential therapeutic interventions.
