The assay follows a six-step protocol: equilibrate reagents and TMB substrate to room temperature and load standards, samples and controls onto the pre-coated plate (37 °C, 90 minutes); add biotin-labelled detection antibody (37 °C, 60 minutes); incubate with HRP-Streptavidin (37 °C, 30 minutes); develop colour with TMB substrate in the dark for 10–20 minutes; then add stop solution and read absorbance at 450 nm. Linearity across 1:2–1:8 dilutions was 87–105% in serum, 84–100% in EDTA plasma and 80–98% in heparin plasma, with mean spike-and-recovery values of 99% (serum), 98% (EDTA plasma) and 96% (heparin plasma).
