Human IL-17A/F ELISpot Kit

The Human IL-17A/F ELISpot Kit from Assay Genie enables enumeration of individual cytokine-secreting cells that release the IL-17A/F heterodimer, a Th17-associated cytokine implicated in autoimmune pathogenesis, mucosal host defense, and chronic inflammatory conditions including psoriasis, rheumatoid arthritis, and inflammatory bowel disease.

Each well of the pre-coated microplate is functionalized with a monoclonal capture antibody specific for the IL-17A/F heterodimer. Upon incubation at 37 °C, secreted cytokine is bound in situ and rendered visible as discrete spots following addition of a biotinylated detection antibody, streptavidin-enzyme conjugate, and chromogenic substrate. Spots can be enumerated manually under a stereomicroscope or with an automated ELISpot reader.

The assay accommodates cell culture supernatants as well as serum samples and supports the stimulation of PBMCs with polyclonal activators such as phorbol 12-myristate 13-acetate (PMA) and ionomycin to drive Th17 polarization. Kit components should be stored refrigerated and protected from light, with repeated freeze-thaw cycles of the conjugate and detection reagents avoided to preserve activity.
