Human IL-17A ELISpot Kit

The Assay Genie Human IL-17A ELISpot Kit enables researchers to detect and quantify individual IL-17A-secreting cells at the single-cell level, supporting characterization of Th17 responses in autoimmune disease, vaccine immunology, and infectious disease research. The kit uses pre-coated PVDF-membrane microplates that capture cytokine footprints released by stimulated human peripheral blood mononuclear cells (PBMCs), with results visualized as discrete spots readable on any standard automated ELISpot reader.

Each kit supplies pre-coated plates, biotinylated detection antibodies, streptavidin-enzyme conjugate, ready-to-use substrate solution, concentrated wash buffer, and lyophilized recombinant IL-17A standard. The recommended sample volume is 100 uL per well, with cell inputs typically ranging from 1 x 10^5 to 5 x 10^5 cells per well depending on the expected secretion frequency. Stimulants such as PMA and ionomycin or antigen-specific peptides are added to the cell suspension during overnight incubation at 37 °C in a humidified 5% CO2 incubator.

For research use only, this ELISpot kit should be stored refrigerated at 4 °C and protected from light; reconstituted standards should be aliquoted and frozen at -20 °C to prevent repeated freeze-thaw cycles. The kit supports up to 96 determinations, providing reproducible quantification of IL-17A-producing cells in cell culture supernatants and serum samples.
