Human IL-1 beta ELISpot Pair

IL-1 beta (IL-1β) is a pro-inflammatory cytokine central to innate immunity and implicated in autoinflammatory and autoimmune disease research. The Assay Genie ELISpot pair supports enumeration of individual IL-1 beta-secreting cells on a PVDF-bottomed microplate, delivering single-cell resolution readouts from human peripheral blood mononuclear cells, splenocytes, and other immune populations stimulated in vitro with mitogens, recall antigens, or pathogen-derived ligands. The matched antibodies are human-specific, with the capture antibody coating the membrane and the detection antibody binding captured cytokine after cell incubation.

The pair is supplied as biotinylated capture and HRP-conjugated detection reagents, used together with a user-supplied ELISpot well plate. Spots are developed with an appropriate substrate and quantified on an automated reader. Standard workflows load 100 uL of cell suspension per well at 100,000 to 500,000 cells per well, producing spot-forming unit (SFU) counts that reflect antigen-specific responder frequency. For reagent handling, aliquot upon receipt, store refrigerated at 4°C, protect from light, and limit freeze-thaw cycles of the detection conjugate.
