The Human IGHG4 ELISA Kit from Assay Genie enables quantitative measurement of human IgG4 (immunoglobulin G4), the least abundant of the four IgG subclasses, normally representing only 3-4% of total IgG in circulation. IgG4 has emerged as a key biomarker in IgG4-related disease (IgG4-RD) research, including studies of autoimmune pancreatitis, Riedel thyroiditis, retroperitoneal fibrosis, and Mikulicz disease. This in vitro assay uses paired monoclonal capture and detection antibodies directed against the human IgG4 Fc region, providing human-specific detection with no significant cross-reactivity with IgG1, IgG2, or IgG3 subclasses. Researchers commonly apply a 50 uL sample volume per determination, with absorbance readings taken using a standard microplate reader. Reagents are supplied in a single kit containing a pre-coated well plate; components should be stored refrigerated, protected from light, and equilibrated to room temperature before each run.
