Human IFN-gamma ELISpot Kit

The Human IFN-gamma ELISpot Kit from Assay Genie is a cell-based immunoassay that quantifies individual interferon-gamma secreting cells, a key readout for antigen-specific T cell immunity. Secreted cytokine is captured locally around each producing cell on a pre-coated PVDF-backed membrane plate, producing a discrete stained spot that an automated reader counts as spot-forming cells (SFC) per well.

Typical applications include vaccine efficacy studies, infectious disease monitoring (tuberculosis, viral, and fungal pathogens), cancer immunotherapy research, and characterization of Th1-type immune responses. The protocol uses a sample volume of 100 uL per well with overnight incubation at 37 °C in a humidified CO2 incubator, followed by detection antibody binding and substrate development.

For best results, store the pre-coated plate sealed in its foil pouch with desiccant under refrigerated conditions and protect detection reagents from light. Avoid repeated freeze-thaw cycles of the streptavidin-enzyme conjugate. Each kit contains pre-coated plate, capture and biotinylated detection antibodies, enzyme conjugate, substrate, wash buffer, and diluents sufficient for one microplate run.
