Description

A colorimetric Sandwich ELISA from Assay Genie, designed for the quantitative measurement of human Isocitrate Dehydrogenase (IDH) in research samples. The kit is intended for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is reactive with human analytes. The detection method relies on a sandwich double-antibody format with biotin-labeled detection antibody, HRP-Streptavidin (SABC), and TMB substrate, with absorbance read at 450 nm after the addition of stop solution.

Assay performance characteristics reported by Assay Genie include a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. Linearity (1:2, 1:4, 1:8 dilutions; n = 5) spans 88–99% / 88–95% / 87–97% for serum, 85–101% / 84–101% / 91–100% for EDTA plasma, and 88–93% / 89–95% / 83–94% for heparin plasma. Spike-and-recovery (n = 5) is 88–103% (average 97%) for serum, 86–96% (average 91%) for EDTA plasma, and 89–104% (average 95%) for heparin plasma. The kit is stored at 2-8°C for 12 months.

The assay procedure includes reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, and color development with TMB in the dark for 10–20 minutes. IDH is highlighted as a key enzyme in the citric acid cycle, with mutations linked to gliomas and acute myeloid leukemia, making the kit relevant to cancer research, biomarker studies, and personalized medicine approaches.
