Description

A colorimetric Sandwich ELISA from Assay Genie is intended for the quantitative measurement of human ICAM-4 (Intercellular Adhesion Molecule 4, also known as LW, CD242 and Landsteiner-Wiener blood group glycoprotein) in serum, plasma, cell culture supernatant, and cell or tissue lysate samples.

The double-antibody sandwich format is reported by the manufacturer with a sensitivity of 1.875 ng/mL and a measuring range of 3.125–200 ng/mL. Assay Genie's linearity data across 1:2 to 1:8 dilutions span 85-104% in serum, 85-101% in EDTA plasma and 83-99% in heparin plasma, while spike-and-recovery averages are 92% in serum and 98% in both EDTA and heparin plasma.

The protocol involves equilibrating reagents to room temperature, loading standards and samples onto the pre-coated plate, incubating at 37°C for 90 minutes, adding a biotinylated detection antibody with a 60-minute incubation, binding HRP-Streptavidin for 30 minutes, developing with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding stop solution. Sample preparation guidance covers serum (clot, 1000 × g for 20 minutes), plasma (anticoagulant tubes, 1000 × g for 15 minutes at 2–8°C), tissue homogenates in PBS with protease inhibitors, cell culture supernatant (2500 rpm for 5 minutes) and cell lysates in lysis buffer with protease inhibitors. Reagents should be stored at 2-8°C for 12 months, and the kit is positioned for cell biology research into ICAM-4's role in cell-cell adhesion, immune response modulation, inflammatory disorders and cancer.
