Human IA2/PTPRN ELISA Kit

Assay Genie's sandwich ELISA is a highly sensitive and specific assay designed for the accurate detection of IA2 (PTPRN) levels in human samples such as serum, plasma, and cell culture supernatants. The kit is intended for researchers studying autoimmune diseases such as type 1 diabetes, where IA2 is recognised as an important autoantigen. IA2 (PTPRN) is a protein tyrosine phosphatase receptor involved in the regulation of insulin secretion and immune responses, and changes in its levels have been linked to the development and progression of autoimmune diseases, making it a valuable biomarker for monitoring disease activity.

The assay uses a colorimetric detection method in a double-antibody sandwich ELISA format. Sensitivity is reported as 0.188 ng/mL with an assay range of 0.313–20 ng/mL. Accepted sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit should be stored at 2–8°C and remains stable for 12 months under those conditions.

Validated linearity for serum (n=5) spans 86–100% across 1:2, 1:4 and 1:8 dilutions, with EDTA plasma (n=5) at 88–105% and heparin plasma (n=5) at 88–99%. Recovery ranges are 85–102% in serum (average 95%), 87–99% in EDTA plasma (average 96%), and 85–104% in heparin plasma (average 92%). The procedure involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after the stop solution is added.
