The HUS1 checkpoint protein is a core subunit of the Rad9-Hus1-Rad1 sliding clamp, also called the 9-1-1 complex, which is loaded onto chromatin at stalled replication forks and DNA double-strand breaks in human cells. This kit from Assay Genie provides the reagents a lab needs to quantify endogenous human HUS1 protein in research specimens, supporting experiments in DNA damage response signaling, checkpoint activation studies, and oncology biomarker work, including work in colorectal carcinoma and chronic lymphocytic leukemia where HUS1 expression has been investigated.

The kit ships with pre-coated microwell strips and is configured as a complete immunoassay package, providing 96 Assays of capacity per box. Researchers run serial dilutions of a recombinant standard alongside unknown samples and read absorbance on a standard plate reader. The workflow is compatible with in vitro cell lysate preparations and with clarified tissue homogenates from cultured human cell lines or patient-derived biopsy material.

Because HUS1 acts upstream of ATR-Chk1 signaling, the kit pairs naturally with western blot analysis of phosphorylated Chk1 and of Rad17, allowing cross-validation of checkpoint activation across the same experimental panel. Each pack of 96 Assays supports comparative analysis across multiple treatment conditions, genetic backgrounds, or matched tumor and adjacent-normal tissue pairs.
