The assay workflow is a sequential procedure: reagents and TMB substrate are equilibrated to room temperature before standards, samples, and controls are loaded; the plate is incubated at 37°C for 90 minutes for primary antigen binding, followed by 60 minutes with a biotin-labelled detection antibody, 30 minutes with HRP-Streptavidin, then 10-20 minutes of colour development with TMB in the dark; absorbance is read at 450 nm immediately after adding the stop solution.
