HSPA5, also known as GRP78/BiP and referred to by synonyms including GRP78, Glucose Regulated Protein 78, BIP, GRP-78, HSP70-5, and MIF2, is a critical protein involved in protein folding and endoplasmic reticulum stress response. It serves as a key regulator of cellular homeostasis and is implicated in cancer, diabetes, and neurodegenerative disorders. This Sandwich ELISA Kit from Assay Genie is specifically designed for the accurate measurement of HSPA5 levels in human samples and is reactive with human specimens.

The kit employs a double-antibody sandwich format with colorimetric detection via TMB substrate, with absorbance read at 450 nm. It accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.375 ng/mL and the measuring range is 0.625–40 ng/mL. Storage is at 2-8°C for 12 months.

Linearity (n = 5) at 1:2, 1:4, and 1:8 dilutions: serum 97-105%, 87-104%, 90-102%; EDTA plasma 84-92%, 89-99%, 82-100%; heparin plasma 82-95%, 80-98%, 80-96%. Recovery (n = 5): serum range 86-102%, average 94%; EDTA plasma range 86-98%, average 93%; heparin plasma range 89-104%, average 97%. The assay procedure includes reagent equilibration to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody for 60 minutes at 37°C, HRP-Streptavidin (SABC) for 30 minutes, TMB development in the dark for 10–20 minutes, and stop solution addition before absorbance reading at 450 nm.
