HSPB1 (UniProt P04792) is a member of the small heat shock protein family (sHsps, 15-30 kDa) and assembles into large, dynamic oligomers that dissociate into smaller, chaperone-active units upon phosphorylation at three conserved serine residues — Ser15, Ser78, and Ser82 — by MAPKAP-K2 (MK2) downstream of the p38 MAPK cascade. In its phosphorylated state, HSPB1 binds F-actin stress fibers and focal adhesion contacts, regulates cell migration and mechanotransduction, and modulates apoptosis through interaction with cytochrome c and caspase-3. Loss-of-function mutations in the HSPB1 gene are causally linked to the peripheral neuropathies Charcot-Marie-Tooth disease type 2F (CMT2F) and distal hereditary motor neuropathy type 2B (dHMN2B). Circulating HSP27 levels have been independently reported as elevated in serum and plasma of patients with non-small cell lung cancer, COPD, end-stage kidney disease, and following coronary artery bypass grafting, supporting its use as a quantitative biomarker in translational research. Each Assay Genie 96-well microplate kit contains pre-coated capture wells, biotinylated detection antibody, streptavidin-HRP conjugate, TMB substrate, stop solution, wash buffer, and a recombinant HSP27 standard calibrated for the standard curve; reconstituted reagents are typically stored at 2-8 °C and frozen at -20 °C for long-term use, with repeated freeze-thaw cycles avoided by aliquoting working solutions. The assay is compatible with serum, plasma, cell lysates, and tissue extracts, and shows no cross-reactivity with murine Hsp25 or αB-crystallin. For research use only.
