Human HSF2 ELISA Kit

This sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of Heat Shock Factor 2 (HSF2) in human samples, with reactivity specific to human specimens. HSF2 is a key transcription factor involved in the cellular response to stress, playing a critical role in regulating the expression of heat shock proteins, and its dysregulation has been linked to cancer, neurodegenerative disorders, and metabolic diseases.

The assay uses a colorimetric detection method read at 450 nm and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The protocol involves equilibration of reagents and TMB substrate, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-Streptavidin (SABC) binding for 30 minutes, TMB substrate colour development in the dark for 10–20 minutes, and addition of stop solution before reading. Sample preparation covers serum (centrifuge at 1000 × g for 20 minutes), plasma (1000 × g for 15 minutes at 2–8°C), tissue homogenate, cell culture supernatant (centrifuge at 2500 rpm for 5 minutes), and cell lysate. Spike-and-recovery testing gave an average recovery of 92% in serum (range 85–105%), 93% in EDTA plasma (range 85–99%), and 100% in heparin plasma (range 92–104%).
