Human HOGA1 ELISA Kit

Assay Genie's Human HOGA1 ELISA Kit is a sandwich ELISA employing a double-antibody format for the quantitative measurement of 4-Hydroxy-2-Oxoglutarate Aldolase 1 (HOGA1) in human samples. The kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, and uses a colorimetric detection method with absorbance read at 450 nm.

Reported performance characteristics include a sensitivity of 0.188 ng/mL and a detection range of 0.313–20 ng/mL. Linearity in serum (n=5) spans 92–105% at a 1:2 dilution, 82–98% at 1:4, and 80–98% at 1:8, with comparable values for EDTA and heparin plasma. Spike-and-recovery averages are 98% for serum, 97% for EDTA plasma, and 95% for heparin plasma. HOGA1 functions as a key enzyme in lysine catabolism, and its dysregulation has been linked to metabolic disorders such as hyperlysinemia, making it a useful biomarker for related research.

The assay procedure involves reagent equilibration, primary incubation at 37°C for 90 minutes, biotinylated detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction read at 450 nm. Kit components include lyophilised standard, biotin-labelled antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer (25X), plate sealer, and a technical manual. Storage is at 2–8°C for 12 months.
