The HMGCLL1 enzyme (also called 3-hydroxymethyl-3-methylglutaryl-CoA lyase, cytoplasmic) catalyzes the final step of leucine catabolism outside the mitochondrion and supports cytosolic ketogenesis, distinguishing it from the mitochondrial HMGCL isoform that drives classical hepatic ketone body production.

Assay Genie provides this research-use immunoassay to support quantitative measurement of the cytoplasmic enzyme in human blood-derived samples, enabling studies of branched-chain amino acid turnover, organic aciduria research, and cancer metabolism where altered ketogenesis has been implicated. The capture antibody is raised against the cytoplasmic HMGCLL1 sequence and shows no significant cross-reactivity with the mitochondrial HMGCL paralogue, allowing isoform-resolved quantification.

The microplate layout accommodates 96 individual determinations per run, and the chromogenic readout can be quantified on a standard absorbance reader. Reconstituted standard and detection antibody vials should be kept frozen at -20 degrees C and shielded from light to preserve activity over multiple freeze-thaw cycles, while unused strip wells remain refrigerated at 4 degrees C inside the sealed pouch with desiccant.

This in vitro research tool supports metabolic biomarker discovery, hepatic function studies, and oncology investigations of HMG-CoA lyase pathway activity.
