Description

From Assay Genie, this sandwich ELISA kit enables quantitative measurement of human HMGB2 (High Mobility Group Box 2), a DNA-binding protein involved in the regulation of gene expression and cellular processes such as inflammation, cancer, and immune response. The double-antibody sandwich format uses colorimetric detection and is reactive with human samples.

Validated sample matrices include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay has a reported sensitivity of 46.875 pg/mL and a standard range of 78.125–5000 pg/mL. Linearity across 1:2, 1:4, and 1:8 dilutions (n=5 per matrix) was 83–100%, 80–95%, and 87–95% for serum; 89–94%, 81–100%, and 85–102% for EDTA plasma; and 86–101%, 86–101%, and 83–95% for heparin plasma. Spike-and-recovery ranged from 87–102% (average 98%) for serum, 95–101% (average 99%) for EDTA plasma, and 93–101% (average 96%) for heparin plasma.

The workflow involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks onto pre-coated wells, and incubating at 37°C for 90 minutes for antigen binding. A biotin-labelled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-streptavidin for 30 minutes. TMB substrate is added and incubated in the dark for 10–20 minutes; after stop solution, absorbance is read at 450 nm. All reagents are stored at 2–8°C, with the kit stable for 12 months under these conditions.
