Human HLA-DRB5/HLA class II histocompatibility , DR beta 5 chain ELISA Kit

Assay Genie's Human HLA-DRB5 ELISA Kit is a competitive ELISA, coated with antigen, designed for the quantitative measurement of HLA-DRB5 (HLA Class II Histocompatibility DR Beta 5 Chain) in human biological samples. The assay offers a sensitivity of 0.938 ng/mL and a working range of 1.563–100 ng/mL, and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit should be stored sealed at 2–8 °C and not cryopreserved.

HLA-DRB5 plays a critical role in antigen presentation and immune response, and variations in the gene have been linked to autoimmune diseases, inflammatory disorders, and infectious diseases, supporting its use as a biomarker in immunology, genetics, and personalised medicine research. Linearity has been demonstrated across 1:2, 1:4 and 1:8 dilutions: serum (n = 10) showed 87–103%, 85–105% and 92–105% respectively, EDTA plasma (n = 10) showed 86–98%, 85–99% and 82–100%, and heparin plasma (n = 10) showed 82–98%, 82–98% and 85–96%. Spike-and-recovery averaged 96% for serum (range 90–102%), 97% for EDTA plasma (range 89–101%) and 94% for heparin plasma (range 86–103%).

The procedure uses a biotin-labelled antibody with TMB substrate and an HRP-streptavidin conjugate; samples and standards are incubated at 37 °C during binding and colour development, and absorbance is read at 450 nm after addition of stop solution.
