Description

From Assay Genie, this Sandwich ELISA kit (double-antibody format) is designed for the precise quantification of Histone Deacetylase 8 (HDAC8) in human samples using a colorimetric detection method, with absorbance read at 450 nm. HDAC8 is a key enzyme involved in the regulation of gene expression through histone deacetylation, playing a role in biological processes including cell growth and differentiation. Dysregulation of HDAC8 has been linked to several diseases such as cancer and neurological disorders, making it a valuable biomarker for research into disease mechanisms and potential therapeutic strategies.

The kit demonstrates a sensitivity of 0.375 ng/mL with a detection range of 0.625-40 ng/mL. It is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity has been confirmed in serum (88-104%, 90-105%, and 86-105% at 1:2, 1:4, and 1:8 dilutions respectively), EDTA plasma (82-99%, 90-99%, and 87-101%), and heparin plasma (82-100%, 84-99%, and 82-99%). Recovery ranges from 85-102% in serum, 86-102% in EDTA plasma, and 86-103% in heparin plasma, with an average of 94% across all three matrices. Reagents should be stored at 2-8°C for 12 months.

The assay workflow includes equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls onto the pre-coated plate, and incubating at 37°C for 90 minutes to allow antigen binding. Biotin-labeled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) binding for 30 minutes. Color is developed with TMB substrate in the dark for 10-20 minutes, the reaction is stopped, and absorbance is measured at 450 nm.
