Description

From Assay Genie, this colorimetric sandwich ELISA uses a double antibody format for the quantification of human Histone Deacetylase 8 (HDAC8). The kit is intended for human samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. According to the manufacturer, HDAC8 is a key enzyme involved in regulating gene expression through histone deacetylation, with dysregulation linked to diseases such as cancer and neurological disorders.

The assay has a sensitivity of 0.375 ng/mL and a detection range of 0.625-40 ng/mL. Linearity across dilutions of 1:2, 1:4, and 1:8 was reported as 88-104%, 90-105%, and 86-105% for serum (n = 5); 82-99%, 90-99%, and 87-101% for EDTA plasma (n = 5); and 82-100%, 84-99%, and 82-99% for heparin plasma (n = 5). Recovery ranged from 85-102% (average 94%) for serum, 86-102% (average 94%) for EDTA plasma, and 86-103% (average 94%) for heparin plasma. Storage is at 2-8°C for 12 months.

The protocol includes reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB substrate color development in the dark for 10–20 minutes, and absorbance reading at 450 nm after the stop reaction. Kit components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody, HRP-Streptavidin conjugate, TMB substrate, sample, antibody and SABC dilution buffers, stop solution, wash buffer, plate sealer, and a technical manual.
