Human Histatin-3 ELISA Kit

Manufactured by Assay Genie, this Sandwich ELISA Kit uses a colorimetric, double-antibody detection method to quantify Histatin-3 in human samples and is intended for research in signal transduction.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay has a reported sensitivity of 7.5 ng/mL and a working range of 12.5-800 ng/mL. Components are stored at 2-8°C for 12 months. According to the manufacturer, Histatin-3 (synonyms: HTN3, PB, Basic histidine-rich protein, Hst, Histidine-rich protein 3) is a protein involved in immune defense and oral health, with antimicrobial properties relevant to studies of periodontal disease and oral candidiasis.

The protocol runs on a pre-coated microplate: standards, samples and controls are incubated at 37°C for 90 minutes, followed by a biotinylated detection antibody (60 minutes at 37°C) and an HRP-streptavidin conjugate (30 minutes at 37°C). TMB substrate is then added and developed in the dark for 10–20 minutes before the reaction is stopped and absorbance read at 450 nm. Reported sample linearity (n=5) at a 1:2 dilution spans 86–102% for serum, 82–101% for EDTA plasma and 80–90% for heparin plasma, with average recovery figures of 90% (serum), 99% (EDTA plasma) and 97% (heparin plasma).
