Description

From Assay Genie, this ELISA Kit is designed for the quantification of heparanase levels in human samples. Heparanase is an enzyme involved in breaking down heparan sulfate, a component of the extracellular matrix, and its dysregulation has been linked to cancer, inflammation, and fibrosis.

The kit employs a sandwich ELISA format with colorimetric detection and uses a double antibody approach. It is reactive with human samples and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, according to the manufacturer. Reported performance includes a sensitivity of 46.875 pg/mL and a working range of 78.125-5000 pg/mL. Linearity for serum (n=5) is given as 82-99% at 1:2, 80-93% at 1:4 and 88-100% at 1:8; for EDTA plasma (n=5) as 81-92%, 84-99% and 86-103%; and for heparin plasma (n=5) as 86-103%, 94-99% and 88-99%. Recovery ranges are 85-99% for serum (average 94%), 89-99% for EDTA plasma (average 95%) and 89-103% for heparin plasma (average 98%). Reagents are stored at 2-8°C for 12 months.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and controls on the pre-coated plate, incubating at 37°C for 90 minutes for antigen binding, adding biotin-labelled detection antibody with a 60-minute incubation at 37°C, adding HRP-Streptavidin (SABC) with a 30-minute incubation at 37°C, developing colour with TMB substrate for 10-20 minutes in the dark, and reading absorbance at 450 nm after adding stop solution.
