Description

A colorimetric Sandwich ELISA from Assay Genie provides quantitative measurement of Human Hemopexin (HPX) in human samples and is supplied for research use only. Hemopexin (synonym: Beta-1B-glycoprotein) is a secreted plasma glycoprotein that binds heme with high affinity and transports it to the liver for breakdown and iron recovery, after which free hemopexin returns to the circulation. Dysregulation of hemopexin has been implicated in hemolytic disorders, liver diseases, and inflammatory conditions.

Reactive with human samples, the assay is validated for serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids, and shows specificity for natural and recombinant human Hemopexin. The target is encoded by the HPX gene (official synonym HX), located at chromosomal region 11p15.5-p15.4, with UniProt accession P02790 and a molecular weight of 51.7 kDa (462 amino acids). Hemopexin is reported to interact with HEV ORF3 protein and FLVCR1, with research applications in Signal Transduction.

Performance characteristics include a detection range of 0.312–20 ng/mL, a sensitivity of 0.078 ng/mL, an intra-assay CV of 5.8%, and an inter-assay CV of 10.3%. Linearity has been evaluated at dilutions of 1:2, 1:4, 1:8, and 1:16 (N=5 per matrix): serum 81–91%, 99–109%, 111–123%, 96–105%; EDTA plasma 94–104%, 95–105%, 94–105%, 81–90%; heparin plasma 83–95%, 81–89%, 108–117%, 98–108%. Recovery averages 103% (range 97–109%) for serum and 105% (range 99–111%) for plasma. Supplied components include an 8×12-strip microplate, lyophilized standard, sample diluent, assay diluents A and B, detection reagents A and B, wash buffer, substrate, stop solution, and plate sealers, with absorbance measured at 450 nm on a microplate reader.
