Description

Assay Genie offers a sandwich ELISA kit for the quantitative detection of human Heat shock cognate 71 kDa protein (HSPA8). The assay is reactive with human samples and accepts serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. Known synonyms for the target include Heat shock 70 kDa protein 8, Lipopolysaccharide-associated protein 1, LAP-1, HSC70, HSP73 and HSPA10, and the kit is specific for natural and recombinant human Heat shock cognate 71 kDa protein (UniProt P11142). Detection is colorimetric.

Reported performance figures include a detection range of 0.312–20 ng/mL and a sensitivity of 0.15 ng/mL. Intra-assay CV is 6.1% and inter-assay CV is 9.9%. Spike-and-recovery averages are 103% (range 97–109%) for serum and 105% (range 99–111%) for plasma. Linearity has been demonstrated across serial dilutions: serum (N=5) showed 99–111% at 1:2, 88–98% at 1:4, 109–119% at 1:8 and 107–119% at 1:16; EDTA plasma showed 100–108%, 102–114%, 105–113% and 102–111% across the same dilutions; heparin plasma showed 91–101%, 112–122%, 87–97% and 86–97%.

HSPA8 functions as a molecular chaperone involved in protein folding, refolding of misfolded proteins and protein complex assembly, acting through cycles of ATP binding, hydrolysis and ADP release mediated by co-chaperones. It is reported in the cytoplasm, melanosome, nucleus, nucleolus and cell membrane, translocating rapidly to the nuclei, and especially to the nucleoli, upon heat shock. The kit is intended for research use only and is associated with the Epigenetics research area.
