Human HCG ELISA Kit

Designed for quantitative measurement of Human Chorionic Gonadotropin (HCG), this Sandwich ELISA from Assay Genie uses a double-antibody format and colorimetric detection. HCG is a hormone produced during pregnancy that plays a crucial role in the maintenance of the embryo and early development of the placenta. Abnormal levels can indicate various health conditions including pregnancy complications, certain types of cancer, and hormonal imbalances, making this kit a useful tool for research in reproductive health, cancer diagnosis, and hormonal disorders.

Reactive with human samples and validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, the assay delivers a sensitivity of 46.875 pg/mL and a working range of 78.125-5000 pg/mL. Dilution linearity at 1:2, 1:4 and 1:8 was reported as 85-95%, 88-99% and 81-87% for serum (n=5); 85-104%, 85-96% and 83-99% for EDTA plasma (n=5); and 86-105%, 89-100% and 87-98% for heparin plasma (n=5). Spiked recovery averaged 94% in serum (range 85-104%), 97% in EDTA plasma (range 90-102%), and 96% in heparin plasma (range 88-104%). Kit components should be stored at 2-8°C for 12 months.

The protocol begins by equilibrating reagents to room temperature, followed by a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody step, a 30-minute HRP-streptavidin step, TMB colour development in the dark for 10-20 minutes, and absorbance reading at 450 nm after adding stop solution. Sample preparation guidance covers serum (clot and centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant collection and centrifuge at 1000 × g for 15 minutes at 2-8°C), tissue homogenate, cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysate preparations.
