Description

A Sandwich ELISA from Assay Genie designed for the quantitative measurement of human Hydroxyacylglutathione Hydrolase (HAGH) in serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. HAGH is described as a key enzyme involved in maintaining cellular redox homeostasis and detoxifying reactive lipid peroxidation products, and its dysregulation has been linked to diabetes, Alzheimer's, and cardiovascular disorders. Detection is colorimetric, based on a double-antibody sandwich format using a biotin-labeled detection antibody and HRP-Streptavidin, with a TMB substrate read at 450 nm.

Reported performance characteristics include a sensitivity of 0.188 ng/mL and a standard range of 0.313–20 ng/mL. Linearity was validated at dilution factors of 1:2, 1:4, and 1:8 for serum (88–103%, 85–97%, 91–99%), EDTA plasma (86–101%, 83–101%, 82–98%), and heparin plasma (82–92%, 85–99%, 80–98%). Spike-and-recovery averages are 97% for serum (range 88–105%), 94% for EDTA plasma (range 85–102%), and 95% for heparin plasma (range 90–99%). The kit is intended for storage at 2–8°C and remains stable for 12 months under those conditions.

The assay procedure follows a sandwich immunoassay workflow: equilibrate reagents and TMB substrate to room temperature, load standards, samples and blanks onto the pre-coated plate and incubate at 37°C for 90 minutes, add biotin-labeled detection antibody (37°C, 60 minutes), add HRP-Streptavidin (SABC; 37°C, 30 minutes), develop with TMB substrate for 10–20 minutes in the dark, add stop solution, and read absorbance at 450 nm immediately. Sample preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate.
