Description

A high-quality sandwich ELISA kit from Assay Genie designed for the quantitative measurement of human 3-hydroxyanthranilate 3,4-dioxygenase (HAAO) in research samples. HAAO is an enzyme involved in the kynurenine pathway of tryptophan catabolism, and its dysregulation has been implicated in inflammatory, neurological and psychiatric conditions. By quantifying HAAO levels, researchers can investigate the role of this enzyme in health and disease and explore potential therapeutic targets.

The assay uses a double-antibody sandwich format with colorimetric detection. Sensitivity is 0.188 ng/mL and the working range is 0.313–20 ng/mL. The kit is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sample linearity has been demonstrated in serum (86–105% at 1:2, 87–103% at 1:4, 85–104% at 1:8), EDTA plasma (83–96% at 1:2, 84–101% at 1:4, 84–101% at 1:8) and heparin plasma (82–95% at 1:2, 81–90% at 1:4, 82–98% at 1:8). Spike-and-recovery performance is reported as 85–102% (average 96%) for serum, 88–99% (average 95%) for EDTA plasma and 86–104% (average 95%) for heparin plasma. Components include a dismountable ELISA microplate, lyophilized standard, biotin-labelled detection antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer and plate sealer; most liquid components are stored at 2–8°C. The protocol proceeds through reagent equilibration, primary incubation at 37°C for 90 minutes, detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition.
