GSTM1 encodes the Mu class 1 isoform of the glutathione S-transferase superfamily, a phase II detoxification enzyme that catalyzes conjugation of reduced glutathione to electrophilic substrates, including environmental carcinogens, chemotherapeutic agents, and lipid peroxidation products. Investigators apply this kit to quantify GSTM1 protein levels in human biospecimens, supporting studies of oxidative stress, xenobiotic clearance, and the clinically important GSTM1 null polymorphism—a homozygous gene deletion carried by approximately half of all individuals that has been associated with increased risk of bladder, lung, and colorectal cancers and altered responses to platinum-based chemotherapy. The Assay Genie format provides a microtitre plate with pre-coated capture antibody, biotinylated detection reagent, recombinant standards, and substrate solution. Each well uses 100 µL of sample, with absorbance read at the appropriate wavelength after stopping the enzymatic reaction. Unopened components should be refrigerated at 4°C, while reconstituted standards and diluted detection antibody should be aliquoted to limit freeze-thaw cycles and held at -20°C for extended stability. The assay is intended for research use only and validated for cell culture supernatant, plasma, and serum matrices in a laboratory in vitro workflow.
